Jessie Hampton was a police officer with the Los Angeles Police Department. She worked at the Van Nuys Precinct and her partner was Officer Phillips. Hampton spoke Spanish fluently.

Shortly after 3am, Jessie Hamtpon heard a shooting at 18166 San Fernando Road and reported it. She got out of her vehicle and began to chase after the assailant, when another armed men appeared. The man identified himself as Jack Bauer with the CTU Los Angeles. Hampton checked his ID and told him she never heard of the Counter Terrorist Unit.
The two of them pursued Greg Penticoff and Hampton turned off her radio. They were forced to take cover from Penticoff when Bauer's cell phone began to ring. When a janitor almost got caught in the crossfire, Hampton spoke Spanish to him and told him to wait until she got back. Hampton was eventually taken hostage by Penticoff and Jack tried to rescue her, but she was killed. Bauer took Penticoff into custody, but Sergeant Kiley was forced to bring Penticoff to the precinct in Van Nuys, as he murdered a cop.
Shortly after 4:30am, Bauer broke Penticoff out of the precinct, as Penticoff knew information that regarded Bauer's missing daughter. After Bauer no longer needed him, George Mason had Penticoff returned to the precinct. It is assumed that he was prosecuted for the murder of Hampton, who was a good cop according to her colleagues.

- The DVD menu for the special edition of Season 1 misspells her name as "Hamilton." In the special edition DVD Hampton appears in several deleted scenes:
- In one deleted scene Day 1 3:00am-4:00am, Hampton tells Jack about the time David Palmer came to her school and made an impression.
- Another deleted scene shows Hampton pleading with Jack to keep Palmer alive, then dying just before being loaded into the ambulance.
